The aim of the work was to study the relationship between some biochemical parameters of the smooth muscle fibers in the corpus cavernosum of the human penis, the histochemical characteristics of the muscular tissue, and the methods of fixation used. A study of the corpus cavernosum of the human penis was performed using fixative and histochemical techniques. The following fixation methods were used: the impregnation by osmium-ferrous-hematoxylin; osmium-hematoxylin-eosin; routine-hematoxylin-eosin-safran; routine-hematoxylin-safran. The cells under study also were stained for SDH, ASAT and LDH histochemically. The impregnation by osmium-ferrous-hematoxylin and the routine-hematoxylin-eosin-safran were not suitable for the study of the smooth muscle cells due to the following: the haematoxylin impregnation produces an intense nucleus staining that contrasts with the impregnation of the cytoplasm by osmium-ferrous-hematoxylin, and the safran reaction is insufficient. The routine-hematoxylin-safran method is the most suitable method of fixation for this kind of study because it allows a better
